E. coli Model for Protein Amplification
Escherichia coli is a bacterium that most of us know first from recalls of foods like meat and lettuce. Although E. coli is a commensal (non-harmful) gut bacterium, the wrong circumstance and strain can make infection uncomfortable and dangerous. E. coli is gram negative, meaning it has a double-layered cell membrane that makes it more difficult to treat with antibiotics.

Our homogenization equipment is regularly used to disrupt E. coli. In âIdentification and Biosynthesis of pro-inflammatory sulfonolipidsâ authors Hou et al. use the NanoGenizer to break apart E. coli after using it for protein expression. Many other researchers have done the same (https://www.genizer.com/u_file/2206/file/6bffa6a456.pdf, https://www.genizer.com/u_file/2312/file/298f8a65e6.pdf ,
https://www.genizer.com/u_file/2312/file/3ca3c35d8a.pdf)

What makes this common cause of diarrheal illnesses a welcome tool in so many research labs?

First, we need to understand the research problem E. coli is being used to address in many labs.

Researchers often need large amounts of proteins for a variety of purposes. One common example is for pharmaceutical production at a large scale. However, having to obtain that protein from its natural source can be time-consuming, resource intensive and complicated. Instead, researchers will often take the gene that encodes that protein and transform it into a bacterium. There, the bacteria will make that protein more quickly. However, not just any bacteria are suited for this purpose, and thatâs what makes E. coli a laboratory star.

Â
Speed: E. coli can double as fast as every 20 minutes. Optimal conditions for quickly producing large quantities of protein.
Â
Density: E. coli can grow densely, reducing the amount of space required for protein production.
Â
Materials: E. coli can grow well on affordable and easily obtainable materials, making it accessible to researchers.
Â
Universality: The wide breadth of research done using E. coli as a model makes it easier to work with.
Â
Tractability: Tractability refers to the ease of editing a genome. E. coli is naturally genetically tractable and can pick up new genes and add them to itself. This eases the process of adjusting its genome for researchers. As a pathogen, this feature also helps E. coli evade antibiotics.
Â
So researchers, go forth and enjoy this model organism in the laboratory, if not always in your kitchen!

The NanoGenizer | A High-Pressure Homogenizer for Laboratory Settings
The NanoGenizer is the most popular unit from Genizer.ÂThis high-pressure homogenizer is perfect for laboratory settings because of its compact and energy efficient design. The NanoGenizer is simple to use. It plugs into the wall and has a convenient touch screen with an auto-stop function. The body of the NanoGenizer serves as the engine to power the ceramic piston. The sample travels in through the inlet valveÂand out the outlet valve into the diamond interaction chamber (DIXC).
The DIXC can be either Y-shaped or Z-shaped. In the Y-shaped interaction chamber, the sample separatesÂinto two streams. The streams turn at a 90Â angle and collide with each other. Where the two streams collide, forces like high shear and cavitation homogenize the sample. While in the Y-shaped interaction chamber the sample collides with itself, in the Z-shaped chamber the sample collides with the wall of the chamber.
In addition to the cooling function in the interaction chambers, Genizer offers a real-time cooling system, with the heat exchanger. This feature is useful for thermally unstable samples. Common applications for the NanoGenizer include but are not limited to nanoemulsions, cell disruption and deagglomeration.

3 Questions to Choose a Liposome Extruder
3 Questions to Ask While Looking for a Liposome Extrusion Option

There are three questions to ask when you are choosing a liposome extrusion option.
First, what pore size do you need? While a solo homogenizer reaches a liposome size of 20-30nm, any option with extrusion allows for more choice.

Second, what power types do you have access to?
The jacketed liposome extruders allow for gas power. An electric option is available if you go for a high-pressure homogenizer with or without an extruder. And manual options are available in the HandExtruder and HandGenizer.

Third, what scale of sample do you need to process? Genizer makes extrusion options from 1 mL to manufacturing scale.

There are advantages coming from the various options as well to help you make your choice!

The difference of High-Pressure Homogenization vs. Sonication
Homogenization is a crucial step in various industrial processes, including pharmaceuticals, food, and cosmetics. High-pressure homogenization (HPH) and sonication are two common homogenization techniques, but HPH offers several distinct advantages over sonication, especially when used with Genizer’s high-pressure homogenizers.
Genizer high pressure homoGenization

Smaller Particle Sizes

One of the most significant advantages of HPH over sonication is its ability to produce smaller particle sizes. HPH operates at much higher pressures and forces the sample through a narrow valve, resulting in a more intense and efficient homogenization process. This leads to smaller particle sizes, which can improve product quality, increase bioavailability, and enhance downstream processing steps such as filtration or extraction.

Using NanoGenizer experimental microjet high pressure homogenizer to homogenize samples
(Left: before homogenization, Right: after homogenization)

Scalability and Energy Efficiency

Another advantage of HPH over sonication is its scalability and energy efficiency. HPH systems can process larger volumes of material, making it a more efficient and cost-effective option for industrial-scale processing. Additionally, HPH is a more energy-efficient process than sonication, which can result in significant cost savings over time.

Gentler Processing Conditions

Finally, HPH can be a gentler process than sonication, particularly when processing delicate samples like pharmaceuticals or biologics. Sonication generates heat and cavitation, which can damage sensitive compounds and reduce product quality. In contrast, HPH provides more precise control over processing conditions, allowing for a gentler and more consistent homogenization process.

When it comes to high-pressure homogenization, Genizer’s high-pressure homogenizers are among the most reliable and efficient options in the market. With advanced technology and user-friendly designs, Genizer’s homogenizers offer precise control over processing parameters, enabling efficient and gentle processing of various samples.

In conclusion, while sonication can be a useful tool for processing samples, HPH offers several advantages over sonication, including smaller particle sizes, scalability, energy efficiency, and gentler processing conditions. By using Genizer’s high-pressure homogenizers, these advantages can be maximized, making HPH a preferred choice for industrial-scale processing in various fields.

Jacketed Liposome Extruder by Genizer: Solutions for Nano

If you are working in pharmaceutical and gene development and delivery, there is a high chance you have at least heard of a liposome extruder. Genizerâs Jacketed Liposome Extruder is leading the industry in innovation and precision.

Liposome extrusion is mainly done to formulate liposomes and achieve a uniform size distribution. Generating nanoscale liposome formulations, and preparing exosomes and artificial cell membranes are other processes that can be done with a liposome extruder.

Diagram of Liposomes extruding through nano sized pores in polycarbonate membrane.

There are many advantages to utilizing Genizerâs Jacketed Liposome Extruder.

Firstly, Genizerâs Jacketed Liposome Extruders are unique in that they have a built in temperature control unit. Most of our jacketed liposome extruders are designed with jacketed barrels. With the ability to hook a cold water unit to its body, the jacketed liposome extruder can guarantee effective extrusion while working with samples that must maintain a certain temperature. This is particularly important when working with pharmaceuticals and genes because of their need to maintain bioavailability.

Secondly, air-compression using a compressed nitrogen cylinder (or other gas chambers) is what drives the power behind jacketed liposome extruders. This ensures that the given pressure is enough to extrude samples through polycarbonate membranes.

Diagram of different liposome extruder sizes ranging from lab scale to pilot scale.

Lastly, there is a wide range of processing capacities allowing Genizerâs Jacketed Liposome Extruders to work in a wide range of applications and industries. The different models of extruders that differ in sample volume size starting with 2mL and going up to 3L in volume. This makes the extruders suitable for laboratory scale use all the way up to pilot scale use. With the addition of Genizerâs Online Liposome Extruders, formulation can even occur on a production and manufacturing scale.

The NanoGenizer Core Processing UnitNanoGenizer High-Pressure Homogenizer

The NanoGenizer is a laboratory scale high-pressure homogenizer. There are two segments of the NanoGenizer, the power unit and the core processing unit. The core processing unit is where homogenization occurs.

Before homogenization, the user places the sample in the inlet reservoir. Genizer offers a variety of options for your inlet reservoir. You may prefer a smaller or larger inlet reservoir depending upon the size of your sample. In addition to stainless steel, we offer plastic syringes and glass inlet reservoirs. Genizer also offers a variety of outlet reservoirs, where the sample goes after processing. You may prefer to use a jacketed glass cylinder, which you can connect to a laboratory chiller circulator for temperature control. Schematic of the NanoGenizer core processing unit, with the inlet reservoir, high-pressure pump, interaction chamber, heat exchanger and outlet reservoirFor small, valuable samples, the syringe option is best. You can hook a Luer hose up to the outlet valve and place the other end in the inlet reservoir to circulate your sample through the core processing unit multiple times. Most samples need 5-10 passes through the diamond interaction chamber (DIXC) to be fully homogenized. Alternatively, you can place the Luer hose into any outlet receptacle in your lab.

The inlet and outlet valve ensures your sample travels the correct direction through the core processing unit. The inlet valve is a one-way valve that allows the sample to enter but not exit. The outlet valve is also a one-way valve and allows the sample to exit but not enter. The sample travels through the inlet valve, into the-high pressure cylinder, and out the outlet valve into the Diamond Interaction Chamber.

The Diamond Interaction Chamber is where homogenization occurs. The chamber can be either Z-shaped or Y-shaped. In the Y-shaped diamond interaction chamber, the sample splits into two streams. Where the two streams collide, forces like high shear and cavitation homogenize the sample. In the z-type chamber, instead of the sample colliding with itself, it collides with the wall of the chamber. After homogenization, the particles are smaller and more uniform and liquids that would normally be immiscible can be mixed.Diagram of the path of liquid Z-shaped and Y-shaped interaction chambers

Genizer’s Diamond Interaction Chamber can come with a cooling option. Additionally, if you need more cooling properties, Genizer offers a real-time heat exchanger. This method of cooling is superior for thermally unstable samples.
The NanoGenizer offers low minimum volume, low dead volume and temperature control options. Please click here for more information on the NanoGenizer.
